Transgenic Mouse Core Facility
The Transgenic Mouse Core Facility (TMCF) is committed to provide customized, flexible, and comprehensive services to UTMB investigators utilizing genetically engineered mouse models. The services we most commonly provide are generation of transgenic mice, gene targeting, cryopreservation of mouse embryos and sperm, rederivation, breeding, and genotyping. We also offer consultation and assist in grant/animal protocol writing. We are located on the ground floor of MRB, inside the ARC.
To provide comprehensive and flexible support to investigators who engage in genetically engineered mouse model research, and to facilitate the growth of the mouse research and translational research in the UTMB.
Generation of Transgenic Mice
Generation of transgenic mice by microinjection of DNA into the pronucleus of a fertilized mouse oocyte
Genetic engineering of mouse embryonic stem cells, and microinjection of the cells into pre-implantation-stage mouse embryos.
A. Cryopreservation of Mouse Embryos
For each day of cryopreservation requested, we will isolate and cryopreserve 8-cell embryos from as many as fifteen super-ovulated female mice using the slow-cooling method.
B. Cryopreservation of Mouse Sperm
We will cryopreserve sperm from two male mice using the Jackson Lab Sperm Cryopreservation Kit. This service includes an assessment for the ability of cryopreserved sperm to fertilize oocytes when thawed, and three-year storage of the sperm in multiple freezers located at the Jackson Lab (Maine and California). Live mice can be recovered by in vitro fertilization, and shipped to the UTMB directly from the Jackson Lab. An additional fee will be applied for this service.
C. Recovery of Cryopreserved Embryos
To recover live mice from cryopreserved embryos, we will perform embryo transfers of thawed embryos into foster mothers.
Rederivation of Mouse Stocks
We can generate specific-pathogen-free mice by embryo transfer from mice that have been infected with viruses or other pathogenic agents. In order to acquire mice from non-commercial vendors, the Animal Resources Center (ARC) may require quarantine or rederivation. We will super-ovulate multiple females and then isolate the embryos from these females using sterile techniques. We will then transfer these embryos into clean foster mothers. Subsequently the ARC will quarantine and test the foster mothers and their offspring to verify that they are specific-pathogen-free.
